EFFECTS OF THE PHYSICOCHEMICAL FACTORS ON THE LIPOXYGENASE OXIDATION OF LINOLEIC-ACID

Oxidation of linoleic acid by 5-lipoxygenase from potato tubers was studied at different pH-values in the presence of the non-ionic surfactant Lubrol PX, which facilitates UV-monitoring of the process. It is shown that linoleic acid and Lubrol PX form mixed micelles, containing linoleic acid in two major forms with pK(a1) = 6.5 (monomeric form localized in the surface layer) and pK(a2) = 8.5 (dimers localized inside hydrophobic core of the micelle). It explains the rise of pH-optimum of the linoleic acid lipoxygenation upon increase of the initial substrate concentration.
