A MONOCLONAL-ANTIBODY WHICH BINDS A CARBOHYDRATE MOIETY BROADLY EXPRESSED ON RAINBOW-TROUT LEUKOCYTES

A monoclonal antibody, anti-2PBL4X (IgG1), was developed which induces aggregation of trout leucocytes and suppresses in vitro mixed lymphocyte cultures and cytolysis by nonspecific cytotoxic cells. Cellular aggregation and suppression of functional assays is mediated by microgram quantities of the antibody and is dose-dependent. The epitope recognised by anti-2PBL4X is present on approximately 75% of trout peripheral blood cells, 84% of trout anterior kidney cells and 81% of trout thymocytes, as determined by flow cytometry. Western blot analysis of an extract of trout leucocytes reveals that multiple bands are stained by anti-2PBL4X. Digestion of the extract with the endoglycosidase F/N glycosidase-F from Flavobacterium meningosepticum abolishes the staining pattern, indicating that the antibody recognises a broadly expressed N-linked oligosaccharide on trout leucocytes. © 1993 Academic Press.
