A bacterial stain to demonstrate the presence of Staphylococcus aureus in harvested osteomyelitis tissue samples from a research animal model is described. A number of standard methods were studied for speed, reproducibility by relatively inexperienced personnel (workstudy students) and the final product. The improved staining technique is a combination of several protocols: Hucker Conn for crystal violet, high concentrations of iodine, 50:50 acetate:ethanol, and a modified Van Gieson counterstain. The method consistently results in high contrasting detail and good photographic quality of surrounding bony tissues in 6 mum sections of formic acid decalcified, paraffin processed bone.
