摘要
The fiber antigen of adenovirus type 2 has been extensively purified by a five-step procedure comprising Freon extraction, centrifugation on a cushion of RbCl, DEAE-chromatography, isoelectric separation in a pH gradient, and gel filtration on 6% agarose. The final product is homogeneous by analytical ultracentrifugation, polyacrylamide electrophoresis at various pH, immunoelectrophoresis, and electron microscopy. Fiber antigen so prepared has a sedimentation coefficient of 6.1 with a calculated molecular weight of 70,000. Immunodiffusion and hemagglutination revealed two antigenic determinants in the purified fiber, one subgroup specific and the other type specific. The purified fiber did not induce neutralizing antibodies detectable in the plaque assay when it was injected into rabbits. © 1968.
