DETERMINATION OF VITAMIN-K-1 IN CANOLA-OILS BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY WITH MENAQUINONE-4 AS AN INTERNAL STANDARD

摘要
An effective high performance liquid chromatography (HPLC) method for measuring vitamin K-1 in canola oil with menaquinone-4 (MK-4) as an internal standard is described. The reliability of this method was examined in detail. No endogenous MK-4 or K vitamins other than vitamin K-1 were found in canola oil. The oil-soluble vitamin fraction was concentrated by a preliminary enzymatic hydrolysis of triacylglycerides, further purified by a silica SPE cartridge, and then analyzed by reverse-phase HPLC with fluorescence detection after post-column chemical reduction with a zinc dust column. Vitamin K-1 in canola oil was confirmed to occur at the level of nearly 400 mu g/100 g of oil, six samples giving a range between 348 mu g/100 g for a crude oil and 125 mu g/100 g for one commercial retail oil. The exact level of Vitamin K-1 in any one oil might be related to the oil origin, refining, and retail display conditions.
