EFFECT OF CISAPRIDE ON THE MOTOR-ACTIVITY OF THE SPHINCTER OF ODDI IN HUMANS - EVALUATION BY ENDOSCOPIC BILIARY MANOMETRY

Endoscopic manometry was performed to evaluate the effects of cisapride on the motor activity of the sphincter of Oddi (SO) in 13 patients with elevated SO pressure and 6 healthy volunteers. Cisapride was given orally to 7 patients and 3 volunteers at a dosage of 10 mg/d (group 1) and to 6 patients and 3 volunteers at 20 mg/d (group 2). Endoscopic manometry was performed before and 1 and 2 weeks after drug administration. All group 2 patients experienced a significant decrease in SO pressure after 2 weeks, and the pattern of SO contraction waves changed from irregular to regular in half of these patients. The results suggest that high doses of cisapride can both inhibit and stimulate SO motor activity, thus improving motor dysfunction.
