We investigated the presence of lymphocyte activating cytokines in alimentary tract tissues. In adult rats, thymocyte activating interleukin-1 (IL-1)-like cytokine was detected in the tongue, esophagus and proximal stomach, but not more distally in the gastrointestinal tract. Conventional and germ-free rats displayed the same level of bioactivity, indicating that the production is not regulated by exposure to microbial antigens. The investigated factor(s) seems to originate in the squamous epithelium where it might serve as a local growth factor. In 11 out of 12 investigated patients with chronic inflammatory bowel disease (CIBD), extracts of intestinal tissue displayed potent IL-1-like bioactivity, whereas normal controls were negative. Upon HPLC gel chromatography, the activity eluted as a single Mr 17K peak and experiments with neutralizing antibodies verified that the active factor was IL-1-beta. The results indicate that IL-1-beta, which is a pyrogenic and cachectic factor, is a pathogenetic mediator in CIBD.
