AUTOPHOSPHORYLATION OF EPIDERMAL GROWTH-FACTOR RECEPTOR IN CELL-LINE A431 AND SUBLINE A431/1522-4 AT EGF AND TGF-ALPHA INFLUENCE

摘要：

The phenomen of EGF decreased phoshorylation activity was observed in cell lines A431 and A431/1522-4 with TGF-alpha gene introduced under the action of TGF-alpha in 5 mM concentration purified from K-media from A431/1522-4 cell Line. At the same time EGF in the same concentration sufficiently increases the EGF-R autophosphorylation. It is shown that TGF-alpha in the lesser concentrations stimulates the autophosphorylation of receptor. Besides, it was observed that in the cells of A431/1522-4 line with TGF-alpha gene introduced the phoshorylation of EGF-R is slightly decreased in comparison with primary 8431 cell line, that is connected with a total decrease of EGF-R concentration in the cells of A431/1522-4 line. In the TGF-alpha-producing cells the substrate with Mr 105-110 kD was detected, its phosphorylation being substantially increased at TGF-alpha addition.
