摘要
Effective, systematic procedures are available for modifying mobile-phase composition to achieve desired band spacings in HPLC separations, but often a change in separation selectivity is best achieved by changing the column rather than the mobile phase. Changing to a column of a different functional type may maximize selectivity better than trying columns of the same type from different manufacturers. The greatest differences in band spacing occur with C8, phenyl, and cyano columns, but because of stability limitations, these columns are not widely used in method development. Sterically protected bonded phases can enhance the stability and reproducibility of these columns and can be used with mobile-phase optimization to separate difficult mixtures.
