This chapter discusses starch gel electrophoresis in vertebrates. Starch gel electrophoresis is used successfully in population genetics, systematics, phylogeny, and population structure studies. The chapter presents guidelines for sample and gel preparation and lists the protein stains and electrophoresis buffers commonly used for a variety of vertebrate species. The standard equipment needed for protein electrophoresis consists of a refrigerated centrifuge, a deep freezer (-70°C) or a liquid nitrogen container, a crushed ice machine, a pH meter, an incubator, an electric barrel homogenizer, a power pack, ultraviolet (UV) light source, and storage boxes for 1.5-ml Eppendorf microtubes. Most of the specific electrophoretic equipment (gel molds, electrode trays, cutting plates, gel slicer) can be made in the laboratory from plexiglass sheets and commercially available material. Stain boxes with lids are required. As most reagents used in electrophoresis are highly toxic, all necessary precautions must be taken, and rubber gloves should be worn when handling acrylamide and stains. © 1993, Elsevier Inc.
