An indirect immunogold-silver staining (IGSS) method for the light microscopical detection of leukocyte cell surface antigens in cell smears was developed. Labeling conditions were determined which gave a good morphology, an intense immunostaining and a low background. The lymphocyte subsets enumerated with this method in buffy coat smears of normal peripheral blood were comparable with those found with the alkaline phosphatase- anti alkaline phosphatase (APAAP) method. This IGSS procedure could form a general method for the detection of cell surface antigens in all kinds of cell smears.
