Isolated potato protoplasts were inoculated with potato Y potyvirus (PVY) particles by polyethylene glycol (PEG). Infection of protoplasts was determined by enzyme-linked immunosorbent assay (ELISA) using anti-PVY antibodies 1, 24 and 48 h post-inoculation (p.i.). 48 h p.i., virus replication was supported by extreme resistant potato cultivars as effectively as by a susceptible cultivar. The extreme resistance to PVY of cultivars bearing the Ry-gene from Solanum stoloniferum was, therefore, not expressed by inoculated protoplasts.
