Hydrogenase (ferredoxin:H** plus oxidoreductase, EC 1. 18. 3. 1) from Desulfovibrio gigas was immobilized by covalent crosslinking using bifunctional reagents, with an activity yield after immobilization of 60%. The activity of the enzyme was influenced by pH and salt concentration. The activation energy had the same value for free and immobilized enzyme. Thermal inactivation was studied. This hydrogenase is relatively resistant to heat inactivation; activity was not reduced after heating at 60 degree C for 1 h. The immobilized enzyme was used for continuous production of hydrogen.
