摘要
The review summarizes the data on the kinetics of plasminogen activation by plasminogen activators (PA) of the tissue and the urokinase types; analyzes the mechanisms of this interaction in the presence of fibrin; demonstrates the regulatory role of the latter in plasminogen activation through direct interaction with the tissue-type PA and indirect interaction with the urokinase-type PA; reveals the functions of these PA and presents experimental and clinical data on their mutually complementary action in eliminating thrombi; establishes the criteria of PA thrombolytic efficacy; assesses the new fibrinolytics derived by chemical modification, recombinant DNA technology, or combinations thereof; demonstrates the prospects of new-generation PA and the importance of combined thrombolytics; and considers the results of molecular, physiological, and clinical studies concerned with PA therapy.
