A CONVENIENT, NONINVASIVE METHOD FOR MEASURING THE KINETICS OF PERMEATION OF DIMETHYL-SULFOXIDE INTO ISOLATED CORNEAS USING NMR-SPECTROSCOPY

摘要
Data for the kinetics of influx of cryoprotectants into tissues is essential for ensuring equilibration and for designing optimised procedures for cryopreservation. Proton nuclear magnetic resonance spectroscopy is used as a non invasive method for assessing the permeation of dimethyl sulphoxide (Me2SO) into isolated rabbit corneas at 0-degrees-C. Corneas reached 64% equilibration within 10 minutes and 82% after 2 hours. Permeation was significantly faster in de-epithelialised corneas.
