摘要
Alkaline phosphatase is determined amperometrically using a simple two-electrode system; results are presented as a kinetic increase in electroactive product. Of the substrates investigated, 4-aminophenyl phosphate gives rise to the optimum signal against background interference in serum. The results are correlated with a routine spectrophotometric assay. The final method is unaffected by interference or fouling problems, with electrodes covered by either a non-selective dialysis membrane or a selective cellulose acetate membrane. Diffusion through both membranes is sufficiently fast for following the kinetic response.
