摘要
The two-hybrid system is a genetic approach to detect the interaction of proteins by expression in a yeast cell. This technique has found wide use in assaying known proteins for interaction and for searching proteins encoded by gene libraries to identify those that interact with a protein of interest. In particular, discoveries resulting from the application of the two-hybrid system to signal transduction and cell cycle control have led to a more complete understanding of these cellular processes. In this article, we present a didactic outline of the two-hybrid system and its application to searching libraries for interacting proteins. (C) 1996 Wiley-Liss, Inc.
