ATP4A联合ATP4B可作为胃癌早期诊断及判断预后的生物标志物

摘要
目的 利用生物信息学筛选出潜在的具有早期诊断及判断胃癌预后的生物标志物。方法 应用GEO2R从GEO数据集筛选出差异表达基因,应用GO/KEGG进行功能富集分析,使用STRING工具生成蛋白互作网络,再利用Cytoscape的MCODE插件鉴定出Hub基因。使用HPA分析Hub基因在胃癌组织中的表达情况,基于Kaplan-Meier分析预后生存曲线。结果 从GSE13911、GSE19826、GSE29272三个数据集得到105个有交集的DEGs,它们主要参与代谢途径。筛选出蛋白互作节点共75个,最后得到10个Hub基因,其中发现ATP4A和ATP4B有明显相关性(r=0.699,P<0.001),且发现ATP4A和ATP4B在胃癌组织细胞浆中表达升高,与预后差呈正相关。结论 ATP4A和ATP4B可能作为胃癌潜在的早期诊断及判断预后的生物标志物。
