A simple and small-scale hydroponic culture system for obtaining second-stage juveniles of the root-knot nematode

We report use of a simple, small-scale hydroponic culture system, using a dwarf variety of mini-tomato, to obtain second-stage juveniles (J2s) of Meloidogyneincognita (Kofoid et White). The system consists of a tomato plant, commercially available hydroponic culture medium, a 500-mL glass beaker, and an airstone connected to an air pump for aeration. The results show that a daily yield of more than 5000 J2s per plant can be obtained for at least for 20 days. Maximum yields of approximately 51,000–66,000 J2s per day were obtained between 11 and 21 days after initiation of the hydroponic culture. The infectivity of the J2s obtained was equivalent to that of J2s collected from egg masses.
