A Rapid and Low Cost Method of DNA Isolation for Marker Assisted Selection in Rice

A simple, rapid, and efficient method of leaf DNA extraction was optimized for PCR based Marker Assisted Selection (MAS) in rice. The procedure involved homogenization of leaves of 10-days old seedlings in the extraction buffer, followed by centrifugation. The supernatant was directly used for PCR. The isolated DNA had protein and RNA impurities. However, these impurities did not affect PCR amplification. Results of comparative analysis of PCR products using the DNA extracted by this method with that using standard CTAB method clearly revealed that the intensity, sharpness, and pattern of PCR amplification were similar. The low cost and time saving method reported here is highly suitable for large scale MAS in rice. © 2014 NAAS (National Academy of Agricultural Sciences).
