Isolation and characterization of fourteen polymorphic tetranucleotide microsatellites for Shangcheng Stout Salamander (Pachyhynobius shangchengensis)

We developed 14 highly polymorphic tetranucleotide microsatellites for the Shangcheng Stout Salamander, Pachyhynobius shangchengensis, a salamandrid endemic to the Dabie Mountain in China. Loci were isolated from a genomic library enriched for (ATAG)n repetitive elements and primers were tested on 21–47 individuals. The number of alleles ranged from 5 to 19 per locus, and the observed heterozygosity (HO) ranged from 0.738 to 0.912. These markers could be a useful tool to investigate population genetic structure, levels of gene flow, population differentiation, mating systems, as well as in studies of landscape genetics in Pachyhynobius shangchengensis.
