Development of 30 SNP markers for the endangered plant Taihangia rupestris based on transcriptome database and high resolution melting analysis

Taihangia rupestris is an endangered plant species, which is disjunctively and sporadically distributed on cliff faces in the southern part of Taihang Moutains, China. Based on the transcriptome sequence data of T. rupestris, 30 single nucleotide polymorphisms (SNP) were developed and characterized in 46 individuals from two populations using high resolution melting analysis. In Lianggou population, the observed heterozygosity ranged from 0.1000 to 0.7727, while expected heterozygosity varied from 0.3717 to 0.5671. For Guanshan population, the observed and expected heterozygosity ranged from 0.0952 to 0.7273 and from 0.3044 to 0.5135, respectively. After Bonferroni correction, two loci in LG population and one locus in GS population showed significant deviation from Hardy–Weinberg equilibrium. These SNP markers will provide useful tools in studying genetic diversity and population structure for the further conservation and management of this endangered plant species.
