New Approaches in Cognitive Neurobiology: Techniques for Two-Photon Vivo Imaging of Cognitively Active Neurons

摘要
This review is focused on the modern methods of in vivo two-photon imaging of activity of neurons involved in cognitive processes in experimental animals. We describe main principles of in vivo two photon microscopy and optical imaging of neuronal activity with two types of calcium sensors: chemical dyes and genetically encoded calcium indicators. We also review a novel approach of in vivo imaging of genomic responses of neocortical neurons to cognitive stimuli using fluorescent reporters for expression of immediate early genes c-fos, Arc and Egr-1 in transgenic mice. We next compare the main advantages and limitations of these approaches and discuss their perspectives for the study of cellular bases of higher brain functions.
