PAR-2 expression and lymphocyte proliferative response inhibition by trypsin

Trypsin has been involved in tumor growth and metastasis. These events are partially mediated by PAR-2 activation on tumor cells. The aim of this work is to evaluate lymphocyte PAR-2 expression and the effect of trypsin on lymphocyte proliferation, IL-2Ralpha expression, and cytokine levels. Trypsin inhibited lymphocyte proliferative-response, IL-2Ralpha expression and IL-2, IL-4, IFN-gamma and IL-10 supernatants levels on PHA-stimulated lymphocytes. In contrast, intracellular cytokines positive cells were not diminished by trypsin incubation. Lymphocyte PAR-2 increased upon PHA stimulation. Our results support that inhibition of lymphocyte proliferation may be induced by cytokines and IL-2Ralpha clevage, suggesting that trypsin may contribute to immune evasion, while promoting growth tumor and metastasis.
