Objective: Objectives: To investigate the expression of GKN1 in gastric cancer and its association with clinicopathological and prognostic significance. Methods: 70 cases of gastric cancer pathological paraffin samples from August 2016 to September 2017 in our hospital for radical gastrectomy were selected retrospectively. Another 40 cases of normal gastric mucosa after gastroscopy were performed from August 2016 to September 2017. Immunohistochemistry was used to stain and record the expression level of GKN1 protein, and to analyze the relationship between GKN1 expression and clinicopathological features in gastric cancer patients. The Kaplan-Meier analysis method was used to map the survival curves of patients with different GKN1 expression levels, and the COX proportional risk regression analysis model was established to explore the indicators affecting the prognosis of patients. Results: The results of immunohistochemistry showed that the expression of GKN1 protein in normal gastric mucosa was significantly higher than that in gastric cancer (P<0.01). The expression level of NEK2 in TNM stage III/IV gastric cancer tissues was significantly lower than that in stage I/II (P<0.05). The expression level of NEK2 in diffuse gastric cancer tissues was significantly lower than that in intestinal type gastric cancer (P<0.05). The expression level in HP-positive gastric cancer tissues was significantly lower than that in HP-negative gastric cancer specimens (P<0.05). Kaplan-Meier univariate analysis of survival curves showed that the 5-year survival rate of patients with low expression of GKN1 protein was significantly lower than that of patients with high expression of GKN1 protein (P<0.05). COX multivariate regression analysis showed that low expression of GKN1 protein was an independent risk factor for the prognosis of gastric cancer patients (P<0.05). Conclusion: NEK2 expression has a certain correlation with TNM staging, Lauren classification and HP infection. The down-regulation or loss of GKN1 expression in gastric cancer is closely related to the occurrence and development of gastric cancer, and could be used as an independent risk factor for the prognosis of gastric cancer patients.
