Disulfanyl peptide decreases melanin synthesis via receptor-mediated ERK activation and the subsequent downregulation of MITF and tyrosinase

摘要
ObjectiveBioactive peptides are commonly used in cosmeceutical purpose. This study was performed to search for an effective and short hypopigmenting peptide using normal human melanocytes as a screening model. A peptide that exhibits multitarget activities will be a promising peptide. MethodsDepigmenting effects were tested in normal human melanocytes. One peptide was selected, and signalling mechanism was investigated by Western blotting and immunofluorescent microscopic examination. ResultsA novel hypopigmenting peptide (dSHP) has been found to inhibit the production of melanin. This peptide significantly decreases tyrosinase activity but was not effective in a direct in vitro assay. It also induces the prolonged activation of ERK, and subsequently downregulates the levels of MITF. PD98059 abolished the dSHP-induced downregulation of MITF. These findings indicate that the dSHP-induced activation of ERK contributes to a reduced melanin synthesis via the downregulation of MITF. Fluorescent microscopic studies were consistent with such findings. Pertussis toxin reverses the downregulation of MITF, which means that the receptor-mediated ERK activation is involved. Moreover, it was also found that downregulation of MITF was clearly inhibited by lysosomal inhibitor (chloroquine). ConclusionNovel tetrapeptide dSHP reduces the melanin synthesis by a receptor-mediated pathway. Furthermore, dSHP works by ERK activation and key transcription factor MITF degradation. Thus, it may be a good candidate as an effective hypopigmenting cosmetic agent. Resume ObjectifDes peptides bioactifs sont couramment utilises dans les produits cosmetiques. Cette etude a ete realisee pour chercher un peptide hypo-pigmentant court et efficace a l'aide de melanocytes humains normaux en tant que modele de selection. Un peptide qui presente une activite multi-cibles sera un peptide prometteur. MethodesLes effets depigmentants ont ete testes dans des melanocytes humains normaux. Un peptide a ete choisi et le mecanisme de signalisation a ete etudie par Western blot et par l'examen microscopique a immunofluorescence. ResultatsUn peptide nouveau hypo-pigmentant (dSHP) a ete trouve pour inhiber la production de melanine. Ce peptide diminue de maniere significative l'activite tyrosinase, mais n'a pas ete efficace dans un essai in vitro directe. Il induit egalement l'activation prolongee de ERK, puis regule a la baisse les niveaux de MITF. PD98059 abolit la regulation negative de MITF induite par dSHP. Ces resultats indiquent que l'activation induite par dSHP de ERK contribue a une reduction de la synthese de melanine par la regulation a la baisse de MITF. Les etudes microscopiques fluorescentes etaient compatibles avec ces conclusions. La toxine de pertussis inverse la regulation a la baisse de MITF, ce qui signifie que l'activation de ERK mediee par le recepteur y est impliquee. En outre, il a egalement ete constate que la regulation a la baisse de MITF a ete clairement inhibee par l'inhibiteur lysosomale (chloroquine). ConclusionLe nouveau tetrapeptide dSHP reduit la synthese de melanine par une voie mediee par le recepteur. En outre, dSHP fonctionne par l'activation de ERK et la degradation du facteur cle de transcription de MITF. Ainsi, il peut etre un bon candidat en tant qu'agent cosmetique hypo-pigmentant efficace.
