Molecular detection of Crimean-Congo haemorrhagic fever virus in ticks collected from infested livestock populations in a New Endemic Area, South of Iran

ObjectiveCrimean-Congo haemorrhagic fever (CCHF) is a viral zoonotic disease with potentially fatal systemic effects on man. We aimed to determine the presence of CCHF virus among collected ticks from domestic livestock from October 2012 to September 2013. MethodsA total of 1245 hard and soft ticks were collected from naturally infested ruminants in Marvdasht County, Fars Province, south of Iran. Nine tick species and one unidentified species in four disparate genera were detected. A total of 200 ticks were randomly selected and analysed by reverse transcription-polymerase chain reaction (RT-PCR) for the presence of CCHF virus genome. ResultsThe viral genome was detected in 4.5% (9 samples) of the studied tick population. The infected ticks belonged to the species of Hyalomma marginatum, Hyalomma anatolicum and Rhipicephalus sanguineus. The viruses detected in these three tick species were clustered in the same lineage as Matin and SR3 strains in Pakistan and some other Iranian strains. These results indicate that the ticks were wildly infected with a genetically closely related CCHF virus in the region. ConclusionRegular controls and monitoring of livestock to reduce the dispersion of ticks and providing information to those involved in high-risk occupations are urgently required. ObjectifLa fievre hemorragique de Crimee-Congo (FHCC) est une zoonose virale avec des effets systemiques potentiellement mortels chez l'homme. Nous avons cherche a determiner la presence du virus de la FHCC dans des tiques recueillies sur le betail domestique d'octobre 2012 a septembre 2013. Methodes1245 tiques dures et molles ont ete recueillies sur des ruminants infestes naturellement dans le comte de Marvdasht, dans la province de Fars, dans le sud de l'Iran. 9 especes de tiques et une espece non identifiee ont ete detectees dans quatre genres disparates. 200 tiques ont ete choisies au hasard et analysees par la reaction en chaine de la polymerase basee sur la reverse transcriptase (RT-PCR), pour la presence de genome du virus de la FHCC. ResultatsLe genome viral a ete detecte dans 4,5% (9 echantillons) de la population de tiques etudiee. Les tiques infectees appartiennent aux especes Hyalomma marginatum, Hyalomma anatolicum et Rhipicephalus sanguineus. Les virus detectes dans ces trois especes de tiques ont ete regroupes dans la meme lignee que les souches Matin et SR3 du Pakistan et certaines autres souches iraniennes. Ces resultats indiquent que les tiques ont ete infectees en milieu sauvage par un virus de la FHCC etroitement lie genetiquement dans la region. ConclusionDes controles reguliers et le suivi du betail pour reduire la dispersion des tiques et la fourniture d'informations a ceux qui sont impliques dans des professions a haut risque sont requis d'urgence. ObjetivoLa fiebre hemorragica de Crimea-Congo (FHCC) es una enfermedad zoonotica viral con efectos potencialmente fatales para el hombre. Nuestro objetivo era determinar la presencia del virus de la FHCC en garrapatas recolectadas de ganado domestico entre Octubre 2012 y Septiembre 2013. MetodosSe recogieron 1245 garrapatas duras y blandas de rumiantes infestados de forma natural en el condado de Marvdasht, provincia de Fars, sur de Iran. Se detectaron 9 especies de garrapatas y una especie no identificada, de cuatro generos dispares. Se seleccionaron 200 garrapatas al azar y se analizaron mediante RT-PCR en busqueda de la presencia del genoma del virus de la FHCC. ResultadosSe detecto el genoma viral en un 4.5% (9 muestras) de la poblacion de garrapatas estudiadas. Las garrapatas infectadas pertenecian a la especie Hyalomma marginatum, Hyalomma anatolicum y Rhipicephalus sanguineus. Los virus detectados en estas tres especies de garrapatas estaban agrupados dentro del mismo linaje que las cepas Matin y SR3 en Paquistan y otras cepas Iranies. Estos resultados indican que las garrapatas estan infectadas con virus geneticamente cercanos al virus FHCC de la region. ConclusionSe requieren urgentemente controles y monitorizacion regular del ganado, para reducir la dispersion de las garrapatas y proveer informacion a aquellos involucrados en trabajos de alto riesgo.
