A review is given of the cellular processes that lead to the formation of platinum-DNA (deoxyribonucleic acid) adducts at the early events that subsequently transpire. Topics discussed include the mechanism by which platinum drugs enter the cancer cells, modification of cellular DNA by platinum-based anticancer drugs, effects of platinum-DNA adducts on the structure and function of proteins that interact directly with the genome, repair of platinum-damaged DNA, the binding mechanism of proteins to platinum-modified DNA.
