Determination of cyclosporine a by high-performance liquid chromatography with aryl oxalate chemiluminescence detection

摘要
Monitoring of cyclosporine A in 40 whole blood samples from renal transplantation patients by high performance liquid chromatography with aryl oxala,te-sulforhodamine 101 chemiluminescence detection is described. These data were compared with fluorescence polarized immunoassay (Cyclosporine-SP Dynapack(TM) using monoclonal antibody for whole blood), with good results (r = 0.88, y = 0.80 + 5.07). These values are the same as reported by high performance liquid chromatography with photometric detection. By using a solid phase extraction cartridge, cyclosporine A was analyzed within 30 min. The analysis time could be shortened to one tenth that previously reported. The proposed method has the same sensitivity as fluorescence polarized immunoassay.
