Determination of the binding constant between progesterone and its monoclonal antibody using affinity capillary electrophoresis

摘要
The binding constant (Kb) between fluorescein isothiocyanate labeled progesterone (FP) and its monoclonal antibody was determined using the ligand separation mode of affinity capillary electrophoresis with laser induced fluorescence (LTF) detection. Free FP and the complexes were separated by capillary electrophoresis in 3 min after 90 min incubation. The running buffer was 50 mmol/L Tricine with pH 8.4, and separation voltage was 20 kV. The Kb was determined to be 4.4 x 10(7) L/mol using Scatchard plotting method. The influences of incubation time and electrophoresis conditions on the determination were also studied.
