Pharmacological modulation of NF-kappa B and HSP70 in a rat hemorrhagic shock model. An immunohistochemical study

The aim of this study was to evaluate the effects of dimethyl sulfoxide (DMSO), pyrrolidine dithiocarbamate (PDTC) and methylprednisolone (MP) on liver, renal and intestinal expression of NF-kappaB and HSP70 after hemorrhagic shock in rats. Rats were hemorrhaged 10-12 ml/kg of their body weight followed by a 45-60 minutes period of hemorrhagic shock. NF-kappaB tissue expression was significantly reduced in liver, kidney and intestine of rats treated with DMSO, PDTC and MP compared to the tissue expression in organs of untreated rats. A simultaneous significant increase of HSP70 expression was observed in liver, kidney and intestine of rats treated with DMSO, PDTC and MP. These results suggest that DMSO, PDTC and MP may represent three efficient inhibitors of NF-kappaB and inducers of HSP70 with potential antishock effects.
