Zonal heterogeneity of peroxisome proliferation in rat liver

In this review the zonal heterogeneity of distribution of peroxisomes and some of their enzymes in the livers of normal rats and in animals treated with hypolipidemic drugs is presented. After a review of the literature we show our own experimental data, in which morphometry and quantitative immunoelectron microscopy have been used. In addition, novel results obtained by nonradioactive in situ hybridization and by Northern blotting applied to isolated periportal and pericentral hepatocytes are presented. All data show clearly a zonal heterogeneity in the induction of lipid beta-oxidation enzymes and their mRNAs in rat liver with a much stronger response in pericentral (zone 3) than in periportal (zone 1) region of the acinus. Our observations suggest that the differential response of hepatocytes to peroxisome proliferating drugs could be related to the state of differentiation of hepatocytes in the liver acinus.
