Germination and cryopreservation of several cactus species from NE Brazil

摘要
Seed germination of Cereus jamacaru, Discocactus zenthneri, Microchanthus flaviflorus and Pilosocereus gounelli was studied under different temperature and light regimes. Discocactus zenthneri seeds germinated at very low percentages (<5%) in all conditions studied; the positive results of the tetrazolium test suggested a very deep dormancy. The other three species had high germination percentages when incubated at 20-30 degrees C. and also at alternating temperatures of 25/15 degrees C, under photoperiod conditions (maximum percentages obtained 96-100%). Incubation in darkness prevented germination (percentages 0-1%). However, when seeds were subsequently transferred to photoperiod incubation. germination was promoted (90-99%). Seed cryopreservation did not affect the germination of the species studied (Cerens jamacaru. Discocactus zenthneri. Melocactus albicephalus, M. concinnus, M. paucispinus, Micranthocereus flaviflorus and Pilosocereus gounelli).
