摘要
Objective: To study the feasibility of matrine treatment of prostate cancer and the mechanism. Method: To a certain concentration gradient of matrine and PC-3m human prostate cancer cells after incubation, determined by MTT method and flow cytometry instrument to detect the changes of the cell proliferation and apoptosis, RT-PCR detection in the PC-3m osteopontin gene (OPNmRNA) expression level. Result: After the matrine role, PC-3m obvious inhibition of growth, and in time-the concentration dependence; Cells in the flow cytometry instrument apoptosis rate increased; PC-3m OPNmRNA expression positive cells; OPN mRNA expression level significantly lower; Conclusion: Matrine in vitro can effectively inhibit the growth of the PC-3m, the mechanism may be associated with induction of cell apoptosis and reduce the expression of OPNmRNA. Osteopontin (Osteopontin OPN) were a secreted phosphorylated glycoprotein that founded in recent years. It Participate in bone metabolism and hardening of the arteries and anti-infection immune response process and so on [1]. Research shows that in recent years, OPN expression is associated with tumor growth and metastasis [2]. Matrine is a kind of alkaloid of active ingredient which extracted from Chinese traditional medicine. It has extensive pharmacological action. In recent years, study of effects of matrine for other tumor cells reported greater increase, its antitumor activity in vivo and in vitro under great attention. We observed the matrine induced human prostate cancer PC-3m cells apoptosis and the expression of OPN, to study the feasibility of matrine for prostate cancer treatment and mechanism.
