摘要
In this study, alpha-amylase producing Bacillus strain FEB5 was isolated from the soil samples collected from Cukurova University campus area. The isolate was identified as Bacillus pacificus according to 16S rDNA nucleotide sequence analysis. B. pacificus FEB5 alpha-amylase showed its maximum activity at pH 11.0 and 40 degrees C. The enzyme retained all its activity after 15 minutes of pre-incubation at 40 degrees C, but above this temperature, it lost its activity gradually until 70 degrees C, but rapidly at the temperatures above 70 degrees C. FEB5 alpha-amylase reached its highest activity level at 24 hours after inoculation. While the enzyme reached higher activity levels compared to the control group in the presence of SDS and Co; CaCl2, MgCl2, and EDTA showed inhibitory effects on the enzyme. SDS-PAGE and zymogram analysis revealed two a-amylase bands of 16.5 and 8.3 kDa.
