Curcumin Combination with miR-144-3p Suppresses Non-Small Cell Lung Cancer by Regulating Nrf-2/HO-1 In Vitro and Vivo Study

Aim: To discuss the effects and mechanism of miR-144-3p in Curcumin-mediated suppression of NSCLC. Methods: Evaluating miRNA-144-3p and Nrf-2 mRNA expression by RT-qPCR and measuring the Nrf-2 protein expression by WB in A549, H1299 and H460 cell lines. After treated by difference methods, measuring the cell viability by MTT assay and cell apoptosis by flow cytometry in cell experiment; by node mice experiment, evaluating tumor volume and weight and positive apoptosis cell number by TUNEL assay. Measuring the relative proteins expressions by WB assay in vitro and by IHC assay in vivo. Results: The cell proliferation rates of Curcumin and Curcumin+ miRNA groups were significantly suppressed with cell apoptosis significantly increasing (P < 0.05, respectively) and there were significantly differences between Curcumin and Curcumin + miRNA groups (P < 0.01, respectively); the tumor volume and weight of Curcumin and Curcumin+ miRNA groups were significantly down-regulation compared with Mock group with positive cell number increasing (P < 0.01, respectively) in vivo; The Nrf-2 and HO-1 proteins expressions of miRNA and Curcumin+ miRNA were significantly down-regulation (P < 0.01, respectively) and PCNA protein expression of Curcumin and Curcumin+ miRNA group was significantly depressed (P < 0.05, respectively) compared with those of Mock group in vitro and vivo study. Conclusion: miRNA-144-3p down-regulates Nrf-2/HO-1 and enhances Curcumin-mediated suppression of NSCLC.
