Affinity maturation of antibody forms part of immunological dogma, although its regulation is poorly understood. Here, David Tarlinton, Amanda Light, Kenneth Smith, and Gustav J.V. Nossal describe their considerable contribution to the analysis of B-cell responses to antigen, and they propose a model for the operation of the germinal center (GC) based on their dissection of various B-cell sub-populations.
