LNCRNA TUG1 CAN INHIBIT THE PROLIFERATION AND INVASION OF GASTRIC CANCER BY INCREASING MIR-506 EXPRESSION

摘要
Introduction: To investigate whether lncRNA TUG1 can inhibit proliferation and invasion of gastric cancer by increasing the expression of miR-506. Methods: A total of 67 cases of gastric cancer patients admitted to our hospital and 62 cases of healthy subjects undergoing physical examination during the same period were chosen to be research objects, with gastric cancer patients as the research group (RG) and healthy subjects as the control group (CG). Experimental gastric cancer cells were purchased and transfected with lncRNA TUG1 and miR-506 to detect their biological behavior. Clinical significance and prognostic significance of lncRNA TUG1 and miR-506 in gastric cancer were explored. The effects of TUG1 and miR-506 on gastric cancer cells were analyzed, and the relationship between them was verified by dual luciferase reporter. Results: NEAT1 in RG was higher than that in CG (P < 0.05), while miR-506 was lower than that in CG (P < 0.05). Receiver Operating Characteristics (ROC) curve analysis showed TUG1 and miR-506 were detected to have a good diagnosis for gastric cancer. Pearson correlation coefficient analysis showed that TUG1 was negatively correlated with miR-506 expression level in RG. The 3-year overall survival rate was 74.63% in RG. TUG1 in survival group was lower than that in death group (p < 0.05), while miR-506 was higher than that in death group (p < 0.05). Overexpressed TUG1 cells had significantly increased proliferation and invasion abilities, but decreased apoptosis ability (P < 0.05). Inhibition of TUG1 cells led to significantly decreased proliferation and invasion abilities, and increased apoptosis ability (P < 0.05). Overexpressed miR-506 cells had significantly decreased proliferation and invasion abilities, but increased apoptosis ability (P < 0.05). Inhibition of miR-506 cells led to significantly increased proliferation and invasion abilities, and decreased apoptosis ability (P < 0.05). The luciferase activity of TUG1-WT was inhibited by miR-506-mimics (P < 0.05). Conclusion: LncRNA TUG1 is highly expressed in gastric cancer and miR-506 is poorly expressed. Both of them have good diagnostic value for gastric cancer and are closely related to the prognosis of patients. LncRNA TUG1 can inhibit the proliferation and invasion of gastric cancer by increasing the expression of miR-506.
