A sensitive automated immunoassay for estradiol on the LIAISON® analyzer

We have developed an automated chemiluminescence immunoassay for estradiol on the LIAISON (R) analyzer. The assay is comprised of paramagnetic particles coated with polyclonal anti-estradiol, a buffer, and a chemiluminescent estradiol conjugate. Data reduction uses a master curve with 2-point calibration adjustment. Results are reported from 20 to 5500 pg/mL (73-20000 pmol/L) with online dilution. Cross-reactivity to estrone is only 7% and <= 2% to other steroids. Mean recovery of added E2 is 101%. Samples dilute linearly, R=0.96. Intra-assay and inter-assay imprecision are 8% and 15% at 50 pg/mL and <= 10% at 70800 pg/mL. Comparison to a commercial RIA gives LIAISON=1.40*RIA-8.6, R=0.97 (0-2000 pg/mL E2). Thus, the LIAISON (R) Estradiol assay provides rapid, sensitive, and precise measurement of E2 in serum or plasma.
