Kinetic analysis of the reaction catalyzed by bacteriophage T7 RNA polymerase

摘要
The kinetics of the reaction catalyzed by bacteriophage T7 RNA polymerase was analyzed theoretically. A reaction scheme taking into account all major steps of the transcription cycle was proposed. By mathematical modeling, equations were derived describing the reaction rate as a function of the concentrations of initiating and noninitiating NTPs and promoter-containing template. Conditions were established in which the reaction catalyzed by T7 RNA polymerase obeys the Michaelis-Menten equation.
