This study aims to evaluate the efficacy of intra-articular injection of chondroitin sulfate (CS) carried by hydrogel in prevention of osteoarthritis (OA) in adult rabbit models. OA models were created with either a trochlea or condyle defect and a-CD-EG 4400 hydrogel was selected as the CS carrier. There were three groups of rabbits; one injected with CS and the other two with hydrogel and saline respectively. At day 50, the defect in the saline group showed no signs of healing, while a thin and slightly irregular layer of fibrous tissue appeared in the hydrogel group. The CS-hydrogel group showed a thicker layer composed of both hyaline and fibrocartilage. In conclusion, intra-articular injection of CS carried by a-CD-EG 4400 is effective in preventing OA in rabbits by improving the histological properties of the knee joint.
