Development of an Inexpensive, sensitive and green HPLC method for the simultaneous determination of brivaracetam, piracetam and carbamazepine; application to pharmaceuticals and human plasma

Racetams have recently become the most important adjunctive therapy in case of many CNS disorders. The goal of the proposed method is to develop and validate a new green and simple liquid chromatographic system for simultaneously quantifying the novel drug brivaracetam with piracetam and carbamazepine. In this study, These three compounds were assayed using an RP C-18 column, with the application of a mobile phase made out of (70:30v/v) mixture of 1% of triethylamine in water and acetonitrile having a final PH of 6.5 adjusted using orthophosphoric acid, with a flow rate of 0.6 mL/min as well as UV recognition at 215 nm in addition to greenness assessment using three assessment tools, namely the Green Analytical Procedure Index (GAPI), the National Environmental Methods Index (NEMI), and the Analytical Eco-Scale. Symmetric well-isolated peaks in a short chromatographic run with a length of <9 min were obtained. The suggested practice was validated and proven to be eco-friendly with high sensitivity as revealed by the values of the limit of quantification which were 3.7, 2.3 and 1.8 mu g/mL for the piracetam, the brivaracetam and the carbamazepine, respectively. The proposed strategy can be utilized to quantify each of the three drugs in their laboratory-prepared synthetic mixtures, dosage form, as well as quantify both the brivaracetam and the carbamazepine in human plasma where the result outcomes were compared with those obtained from the reference methods.
