Measurement of concentration and viability of brook trout (Salvelinus fontinalis) spermatozoa using computer-aided fluorescent microscopy

The usefulness of computer-aided fluorescent microscopy for measuring the concentration and viability of brook trout spermatozoa was tested with the NucleoCounter SP-100 system (Chemometec, Denmark). Sperm concentration measured with the new method significantly correlated with results obtained with the use of the photometric method (r(2) = 0.96; P < 0.0001; n = 23). A dose-dependent relationship was found between the percentage of killed spermatozoa and viability. Our data suggest that using computer-aided fluorescent microscopy with the employment of NucleoCounter SP-100 offers an accurate and fast measurement of sperm concentration and viability of fish semen.
