摘要
Single molecule imaging allows the observation of molecular processes in living cells and provides insights into the hidden cellular events. It is essential that the concentration is low enough to avoid confusing molecules that come so close to each other for an unambiguous identification in subsequent images for reliable identification of molecule traces. Single molecule detection within live cells with a spatial precision in the nanometer range provides successful analysis of complex intermediate-sized problems. The variability in protein mobility can be related to interactions with intracellular components or macromolecules and organelles, which act as obstacles. Molecules performing specific tasks within the nucleus exhibit a modulated mobility, and deliver detailed information on intranuclear interactions and dynamics.
