Novel D-π-A type near-infrared fluorescent probes for the detection of Aβ40aggregates

摘要
Aberrant accumulation of Amyloid-beta (A beta) peptide is closely related to Alzheimer's disease. Thus, it is important to develop featured probes for the specific detection of A beta species. Herein, we designed and synthesized a novel near-infrared fluorescent probeSDPYbased on the D-pi-A architecture for the detection of A beta aggregates. The probeSDPYdisplayed higher affinity for A beta(40)aggregates over A beta(42)aggregates in solution (K-d= 164 nMvs.2.1 mu M). In addition,SDPYshowed excellent anti-interference against a wide range of other substances. Furthermore,SDPYwas capable of labeling A beta(40)aggregates better than A beta(42)aggregates in the brain sections of AD transgenic mouse models.
