Objective:To investigate the effect of osteoblast of type 2 diabetes mellitus (T2DM) patients on the osteoclast formation. Methods: THP-1 cells were differentiated into multinuclear osteoclasts by pro-resorptive cytokine receptor activator of nuclear factor-kB ligand (RANKL) and M-CSF. The alveolar bone osteoblasts were obtained from T2DM patients and normal patients. The cell viability was tested by alkaline phosphatase (ALP) chemical staining. Osteogenic characteristics were measured by alizarin red staining. The differentiation of THP-1 cells into osteoclast cells induced by co-culturing with the supernatant of T2DM osteoblast and normal osteoblast was measured by Tartrate-resistant acid phosphatase-positive (TRAP) staining. Result: There was no significant difference of morphology between the two form cells under the same culture conditions. THP-1 cells could differentiate into osteoclast cells by co-culturing with the supernatant of osteoblast of T2DM patients and M-SCF. The TRAP-positive multinucleated osteoclast was significantly increased by coculturing with the supernatant of osteoblast of T2DM patients and M-SCF. Conclusions: THP-1 cells can differentiate to osteoclast cells induced by co-culturing with the supernatant of T2DM osteoblast and M-CSF, which may explain the phenomenon that T2DM patients are more susceptible to bone regeneration problem, such as osteoporosis and periodontics.
