Enantioseparation of Duloxetine Hydrochloride by Capillary Electrophoresis with Glycogen as Chiral Selector and Its Separation Mechanism

摘要
The chiral separation of duloxetine hydrochloride enantiomers by capillary electrophoresis was studied with glycogen as a novel chiral selector. A new method for enantioseparation was developed. The influences of the concentration and pH of buffer solution, glycogen concentration in the background electrolyte and separation voltage were investigated. The optimum conditions were obtained with 25 kV as separation voltage, 3.0% (m/V) glycogen in 50. mmol/L Tris-H3PO4 (pH 3.0) as running buffer. Under optimum conditions, baseline separation was achieved between the two enantiomers of duloxetine hydrochloride with the resolution of 1.84. A brief mechanism of chiral separation was also discussed.
