Genetic Diversity of Phytoplasmas Associated with Jujube Witches' Broom Disease in Ziziphus jujuba Mill. 'Dongzao'

Jujube witches' broom disease (JWB) caused by phytoplasma is a destructive disease of Chinese jujube. 'Dongzao' is a popular fresh-eating cultivar but is very sensitive to JWB with a higher occurrence in northern China. The CTAB method was used to extract genomic DNA from diseased 'Dongzao' plants grown in four provinces in northern China. The 16S rRNA gene was cloned and sequenced by polymerase chain reaction (PCR). The homology analysis and virtual AFLP analysis showed that phytoplasmas associated with JWB belonged to 16Sr-V group. Simultaneously, it was also found that phytoplasma associated with JWB disease from Beijing, Hebei, Shandong, Henan had similarity of nucleotide sequences higher than 98.5%. This result means that those phytoplasmas of diseased 'Dongzao' from different geographical distribution may be ascribed to the same strain. So far there has been no report on determining the diversity within same cultivar from different geographical locations.
