摘要
Pou5f1 (Oct4), a key gatekeeper of cellular pluripotency, has little been studied with regard to roles of its regulatory cis-elements in transcriptional control of neighboring genes. In this study, bi-allelic deletion of the Pou5f1 promoter with simultaneous rescue of mouse embryonic stem cell (ESC) self-renewal by a 9.8-kb fragment of Pou5f1 knocked into the Rosa26 locus has been performed. The results confirm the previously established Pou5f1 gene boundaries, whereas the generated ESCs will be indispensable for further studies of the Pou5f1 promoter in transcriptional regulation of neighbor genes.
