摘要
Murine embryonic fibroblasts (MEF) derived from TRAF6 knockout mice have increased sensitivity to actinomycin D (ActD) plus TNF-alpha-induced cell death compared with wild-type MER While ActD plus TNF-alpha-induced phosphorylation of JNK and ERK, and degradation of I kappa B alpha were not different between TRAF6(-/-) and wild-type MEF, slightly higher caspase-3 activity was observed in TRAF6(-/-) MER In addition, reactive oxygen species (ROS) were more accumulated in TRAF6(-/-) MEF than in wild-type MEF. An antioxidant, butylated hydroxyanisole (BHA) markedly inhibited cell death in TRAF6(-/-) MEF We assumed that resistance against ActD plus TNF-alpha- induced cell death in MEFs is rendered by TRAF6, which downregulates ROS production.
